Matrix Assisted Laser Desorption Ionization Mass Spectrometry for the Analysis of Inhomogeneity in Dried Polymer Droplets

A convenient sample preparation method in MALDI mass spectrometry is applying a small droplet of a mixture of sample and matrix onto a steel target and letting it dry (so called dried droplet technique). However, depending on the conditions used to prepare the spots it has been observed that the material does not deposit homogenously but may form more or less pronounced rings1.
We have investigated how different polymers, solvents and temperature influence the formation of these rings and used Laser desorption to image the ion intensities. Also the effect of the scanning protocol of the laser was studied. It has been found that it is beneficial to scan every second spot in every second line and afterwards the remaining spots in between than to simply scan every spot in every row due to increased evaporation of material from neighboring positions2.
