Ca2+ release-activated Ca2+ (CRAC) channels are activated by the physical interaction of the Ca2+ sensor STIM1 in the endoplasmic reticulum and the Ca2+ channel, Orai1, in the plasma membrane. An Orai1 mutation (L138F) associated with tubular aggregate myopathy (TAM) exhibits constitutive activity independent of STIM1, which Yeung et al. [1] employed to provide novel insights into the Orai1 pore opening mechanism and fast Ca2+-dependent inactivation (FCDI).
